レチノイン酸支配下の成長因子ミッドカインの受容体構造と活性阻害

研究概要	新しい成長分化因子ミッドカインの構造と機能の関連について研究した。化学合成によって調製したミッドカインの1/2分子を用いてC末端側の1/2分子が強いヘパリン結合能、神経突起伸長能、そして血管内皮細胞のプラスミノーゲン活性化酵素の活性促進能を持つことを見出した。さらに、バキュロウィルスでミッドカインを大量調製することに成功し、この系を利用してミッドカイン変異体を作成した。C末端1/2分子へのヘパリン結合モチーフの中核となる83番目と84番目のリジン残基をグルタミン残基に変えるとミッドカインの神経突起伸長能は完全に失活した。このことからヘパリン結合ドメインがミッドカインの機能と密接に関連することが明白であり、ヘパリンのどの構造モチーフが重要かを検討した。選択的脱硫酸の結果、2-0,6-0,2-Nの全ての硫酸基が神経突起伸長の阻害に重要なことが判明した。また、ヘパリンオリゴ糖のサイズとしては20糖が必要であった。いっぽう、ミッドカインと結合する膜タンパク質の解析から、分子量53,000のタンパク質がミッドカインのタンパク質性受容体である可能性が浮かび上がってきた。ミッドカイン受容体を送気レベルで検出する手法を案出することも出来た。ミッドカインと発がん、心筋梗塞、脳梗塞、リューマチ性関節炎の関連を示すデータも多く得られ、さらに、臨床的解析に利用出来るミッドカインの高感度検出法も確立した。cDNAトランスフェクション実験により、ミッドカインがNIH3T3細胞をがん化させることも見出した。
