細胞接着反応を阻害するプロテオグリカン、抗一接着分子としての細胞挙動の調整とその機構

研究概要	1)プロテオグリカンPG-Mの抗一細胞接着活性の機構の研究:giycocalfinの関与を予想し、数種類のヒト培養細胞からニワトリの系で確立した方法によりヒトgiycocalfinを精製した。まずニワトリglycocalfinに相当する活性と性質の有無を調べた。Ca^<++>イオン依存性のコンドロイチン硫酸へのアフィニティーを示し、また細胞表面に分布して期待されたglycocalfinの性質を示すと同時に、V8プロテアーゼ断片のアミノ酸配列や免疫学的性質は完全に従来のヒトのannexinVIと一致した。既報のヒトannexinVIのcDNA配列をもとに、PCR法を利用してヒトのglycocalfin/annexinVIの遺伝子を得た。これをもとにglycocalfinのアンチセンス遺伝子、ドメイン部分の遺伝子やその変異遺伝子などの発現系を構築し、細胞に導入して影響をみる実験の準備は整った。 2)PG-Mの抗一細胞接着活性による細胞挙動調整の可能性の研究:PG-Mのコア蛋白質のcDNA解析、mRNAの解析、さらにgenomic遺伝子の解析をヒトとマウスについても展開し完了した。PG-Mは、組織特異的また発生段階特異的にアルタナディブスプライシング機構により1分子当りのコンドロイチン硫酸の結合本数がゼロから最多の14本までに多様に変化して合成されており、この性質によりPG-Mはその抗一細胞接着活性を可変できる分子と推定された。発生初期には、全組織で最多数のコンドロイチン硫酸鎖を持つ分子形を、一方、骨格筋と脳には、鎖を持たない分子形の定常的な発現が見られた。骨肉腫細胞MG63へのアンチセンス遺伝子トランスフェクションによるPG-M発現抑制によって、その抗一接着活性の低下によると思われる種々の細胞挙動(細胞骨格系分布、細胞形態、細胞増殖)の変化が観察された。
