細胞接着を阻害するプロテオグリカン、抗-接着分子による細胞挙動の調整とその機構

研究概要

PG-Mのコンドロイチン硫酸鎖の細胞表面レセプターと予想される新分子、glycocalfin(グリコカルフィン)の遺伝子の単離を目指した。ヒト培養細胞株の数種類についてコンドロイチン硫酸アフィニティーカラムへの特異的吸着を利用して分子量67kDaのglycocalfinを得た。そのV8プロテアーゼ消化断片のアミノ酸配列は、既報のヒトannexinVIのそれと一致した。また、市販のannexinVI抗体と反応し、前年度のニワトリ分子の結果と合わせて、glycocalfinは、既知分子であるannexinVIそのものであると結論した。ヒトannexinVIのcDNAを大阪大学・医学部・祖父江憲治博士から得て、現在、分子生物学的方法による同一性の確認を行っている。PG-Mのコンドロイチン硫酸鎖の細胞接着阻害活性発現におけるglycocalfinの関与を予想させる実験、つまりこの細胞接着阻害活性が細胞内シグナル系を介した反応によるかどうかを以下の実験で調べ、いずれの結果も関与説を支持した。フィブロネクチン基質をコートしたU型培養皿に細胞を播種し接着をみた。PG-M添加すると、細胞は円形を保ったままで伸展しない。しかし、培養皿を遠心しても細胞はU底に集まらなく、確かに基質には接着していると思われた。インテグリンをリポゾームに組み込んだ接着モデル実験で、インテグリン-リポゾーム体のフィブロネクチンへの接着はPG-Mでも何らの影響を受けなかった。さらに、細胞を薄いホルマリン固定液で処理して、インテグリン活性を残してシグナル伝達経路を失活させる条件では、固定細胞はフィブロネクチンへ接着し、PG-M添加の影響を受けなかった。これらから、細胞表面レセプターを介したシグナル伝達により、インテグリンのリガンド結合に伴って誘起される細胞伸展に到る一連の反応経路が遮断される機構が推定された。
