蛋白工学とクライオン電子顕微鏡を用いた分子立体構造に基づく蛋白質モーター機構の研究

研究概要	ディクチオ型粘菌の遺伝子操作により、ミオシン重鎖のN端近傍(Λsp5)にシステインを導入し、ここに金原子クラスターを標識し、その位置をクライオ電子顕微鏡によって三次元的に決定した。この結果をX線結晶解析で決定されたミオシン分子と比較することにより、アクチンと相互作用しているミオシンのN端の三次元的位置を決定することが出来た。ミオシンのN端はアクチンから遠い側にあり、アクカリ軽鎖と近い部位に存在していることの、実験的な根拠が初めて得られた。アクチンのトロポミオシン結合部位は、活性型の細いフィラメントでは、アクチンの内側のドメインにあることが、三次元像再構成法の結果の解釈から推測されてきた。本研究により、アクチンを改変し、このドメインのアミノ酸を荷電アミノ酸と非荷電アミノ酸の間で変化させるとトロポミオシン結合能が変化するが、逆に外側のドメインでこのような変異を導入しても、トロポミオンの結合能には変化が無い。外側のドメインの中でも、第4サブドメインのセリンに富むループ(SRL)の近傍のアミノ酸がトロポミオシン結合にとって、重要であると特定出来た。アクチン・トロポミオシン・トロポニンから再構成した細いフィラメントの三次元構造をCaイオンの有無で比較したとき、Caイオンが存在するとき密度で増える領域と、上記のセリンに富むループ(SRL)とは良く一致するので、この部位をトロポミオン結合部位とすべき根拠はより深まった。Caイオンが存在しない時は、外側のドメントの密度が有意に上昇することは既に確立したが、天然の細いフィラメントでも同様の結果が得られた。
