研究概要

イントロンはタンパク質の部品「モジュール」の境界によく対応していることから、分化進化において、モジュールが「かき混ぜられた」基本構造部品であることが明らかになってきた。本研究の目的はモジュールを分類し、機能の異なるタンパク質間に共有されるモジュールを明らかにすることであった。本研究の成果はモジュールを分類できる見通しが得られ、すでにモジュールの一部分がカタログ化できたことである。モジュール図鑑の電子出版やwwwでの公開も検討している。得られた具体的な成果を以下に述べる。
1.モジュールの分類法の確立:部品として機能するため必要な性質として、モジュールの立体構造は内部の相互作用によって決まることが明らかになった。すなわち、他のモジュールから切り離した単独モジュールが力学的に安定であることを、分子動力学計算の結果から示せた。従って、モジュールの分類は立体構造に基づいて行うのが合理的であると結論できた。
2.モジュールの分類:アミノ酸配列の一致度が40%以下であるタンパク質77個についてモジュールを同定し、得られた950個のモジュールを分類した。モジュールが3個以上含まれるグループが66種類特定された。しかし、どのグループにも属さないモジュールが存在している。
3.タンパク質でのモジュール共有:同じグループに属するモジュールが複数のタンパク質に共通に使われていることが明らかになった。これらのモジュールの中には、機能が明確に規定されるものが多くあり、モジュールのかき混ぜが起きたことが明らかである。ヘリックス・ターン・ヘリックス・モジュールには、異なる機能を持つモジュールへの分化が見られた。
