基本転写因子TFIIAのコアクチベータ-としての機能

研究概要	遺伝子発現の最も重要な段階である転写開始反応に関与する蛋白性因子は、各遺伝子に特異的に働くアチベータ-と全てに共通に働く基本転写因子とに分類されている。基本転写因子には、RNAポリメラーゼII、TFIIA、TFIIB、TFIID、TFIIHなどがあり、それらは転写開始複合体を形成する。近年、それらに属さないコアクチベータ-やメディエーターなどと呼ばれる新たな因子群が見つかった。これら因子の機能は、アクチベータ-からのシグナルを転写開始複合体に伝え、RNAポリメラーゼIIによる転写開始反応を促進すると考えられる。従って、コアクチベータ-の構造と機能の解析は、転写開始反応の調節機構を解明する上で非常に重要な研究課題である。我々は、近年、基本的転写因子の一つであると従来考えられてきたTFIIAが、コアクチベータ-として働く可能性を世界に先駆けて示した。本研究では、TFIIAがコアクチベータ-として機能することをカイコの実験系を用いて確証し、その系でTFIIAと相互作用するMBF2を同定し、そのcDNAをクローン化した。MBF2は、DNA結合性アクチベータ-とTATA結合性タンパク質(TBP)との間を架橋するMBF1と結合し複合体を形成するので、アクチベータ-からのシグナルは、MBF1、MBF2およびTFIIAを経由して転写開始複合体に伝達されると考えられる。この研究を行うに当たり、TFIIAを動物細胞抽出液から回収率よく多量に調製する技術開発として、TBP固定化ラテックス粒子を設計した。また、TFIIAと相互作用するアクチベータ-のいくつかDNA固定化粒子を用いて分離・精製し、それらcDNAを単離した。さらに、ペプチド、単鎖DNA、薬剤などを固定化したラテックス粒子を設計し、それらが極めて優れた機能を有することを示した。
