上皮の遺伝子発現を制御する間充織因子の研究

研究概要	本研究の目的は消化管の発生において上皮の形態形成と遺伝子発現を制御する間充織因子を同定することである。ニワトリ胚消化管は、孵卵6日ごろから活発な形態形成を開始し、食道、胃(前胃と砂嚢)、小腸、大腸などの各器官が明瞭に分化する。ついで各器官の上皮は間充織の作用をうけて固有の遺伝子発現を開始する。例えば、前胃の上皮は腺を形成して腺上皮は前胃に特異的なペプシノゲン遺伝子を、孵卵9にちごろから開始する。このような腺の形成とペプシノゲンの発現の関係を探るために、腺形成のような形態に重要であるといわれるコラーゲンの合成を阻害し、ペプシノゲンの発現に対する影響を調べた。その結果、コラーゲンの合成が阻害されると腺形成は完全に抑制されるが、それでも上皮細胞の内本来腺を形成する細胞はペプシノゲンを発現した。従って、上皮におけるペプシノゲンの発現は、腺形成とは独立した事象であることが結論される。また腺形成に伴って、いくつかの遺伝子や抗原(例えばsonic hedgehog,Sox2,cytokeratin,MA-antigen)の発現が完全に消失したり、発現量が著しく低下することが明らかになった。これらの遺伝子や抗原の挙動の変化はやはり間充織によって影響される。間充織の作用の本性を探るために、前胃間充織の代わりに肺間充織を用いたところ、前基間充織と同等またはそれ以上の誘導活性が示された。またその作用が、上皮の細胞増殖活性を増加させることと結びついていることが示唆された。従って、間充織の作用因子としては、種々の増殖因子を想定することができる。
