侵襲・非侵襲法による霊長類(サル・ヒト)の情動・記憶の大脳辺縁系機構

研究概要	1)侵襲法(神経生理学的研究) サル中隔核(SP)から単一ニューロン活動を記録し,サルの居場所の違いにより呈示物体の意味が変化する条件課題(PGN課題)および呈示物体の意味が常に一定である非条件課題(AGN/SGN課題)に対する応答様式を解析した。その結果,サルSPより430個のニューロン活動を記録し,そのうち190個が何らかの応答を示した。I.これら応答ニューロンの30.5%はサルの居場所に関して識別的に応答した。II.条件(PGN)および非条件(AGN)課題の両課題をテストできたSPニューロンのうち,59.7%はPGN課題に対してだけ,その意味(Go-報酬/Nogo-無報酬)に応じて識別的に応答した。このような条件課題特異的ニューロンは,本研究により初めて見出され,SPが条件性・場所-物体識別課題の遂行に重要な役割を果していることを強く示唆する。III.PGNおよびAGN課題に非識別的に応答したニューロンに対してさらにSGN課題をテストし,ほとんど(90.5%)は物体の報酬性(報酬/無報酬)に対して識別的に応答した。これらニューロンの応答性は,消去学習により物体の報酬性を直接変化させるとそれに応じて変化することからも確認された。以上からSPは,居場所の認知→居場所の違いにより変化する物体の意味の識別という一連の過程を統合する上で重要な役割を果たしていることが示唆される。 2)非侵襲法(双極子追跡法,機能的MRI法) i)パターンオンセット刺激による視覚誘発電位(VEP)の電流発生源(双極子)を双極子追跡(DT)法により推定した。等価双極子は,視野の中心部刺激では,刺激野反対側の後頭葉視覚領の後方に,視野の周辺部刺激では,刺激野反対側の後頭葉視覚領の前方に推定され,非侵襲的にヒトの後頭葉視覚領における網膜部位局在性をほぼ再現することができた。また,顔写真に対するVEPを解析することにより,潜時約150msecで後頭葉-側頭葉移行部に双極子が両側性に存在することが明らかになった。ii)昨年度に開発した特殊脳定位固定装置を用い,リック(舐め)行動中のラットの頭部をIMRIで撮影した。グルコース溶液および蒸留水のリック行動中の信号強度を差分した結果,グルコース溶液摂取により視床下部の信号強度が増加(血流が増加)することが明らかになった。
