研究概要	CD46発現トランスジェニックマウスの作成 CD46のコンストラクトをin vitroにおいてCHO細胞等にトランスフェクションして発現を確認した。ついで、これをマウスの受精卵にインジェクションしてトランスジェニックマウスを作成した。しかし、いずれの場合も発現が見られなかった。原因を追及したところ、膜蛋白質であるCD46の細胞内ドメインを削ったトランスジェニックマウスを作成すると発現が認められることを見いだした。即ちCD46のcDNAの細胞内ドメインまたは、3′非翻訳領域には発現を抑制する活性が隠されていることが判明した。さらに、詳細に検討したところ、3′非翻訳領域を削ることによって作成したトランスジェニックマウスのすべてに発現が認められたので、サイレンサー領域は3′非翻訳領域に局在していることが判明した。種々検討の結果、CD46を発現するマウスを作成することに成功した。 CD46genome遺伝子のクローニング上記CD46発現トランスジェニックマウスの発現パターンをみると筋肉、膵臓、腎臓、肺などで強く血球細胞ではそれほど強い作用は認められなかった。麻疹ウイルスに感染する疾患モデルマウスとして応用するために、麻疹ウイルスの感染実験を行ったが感染したと言う十分な証拠は現在まで得られていない。そこで、原点に立ち戻り、CD46をCD46のプロモーターで発現させるべくCD46遺伝子のプロモーターをゲノムからクローニングした。受精時における働き CD46には麻疹ウイルスのレセプターとしての働きや補体の不活化の他に受精の際の相互認識に役立っていると思われるが、精液中には精子以外に由来するCD46が存在することを証明した。このCD46はprostasomeと呼ばれる顆粒の膜上に存在し、補体の制御活性を持っていることも明らかとなった。射精時に膣内の補体から精子を防御する役割が示唆された。
