微小血管からの酸素放出動態の2次元イメージングに関する研究

研究概要	末梢組織の微小血管ネットワークにおける赤血球の酸素飽和度と赤血球の流動状態を示す二次元画像を作成するために試作された装置を用いて、その応用を試みた。1.本装置(構成:倒立顕微鏡、ビデオカメラ、干渉フィルター、画像処理装置およびコンピュータ)は微小血管を流れる血液の酸素飽和度ならびに赤血球の濃度分布状態の2次元画像を作成することにあるが、詳細な解析のため任意の微小血管断面におけるプロフィルを随時得るようにした。2.本装置は、酸素放出の起こる全ての微小血管領域で、測定精度±10%以内で定量測定が可能であることが確認され、微小血管網を赤血球が細動脈から細静脈に流れる過程で酸素を放出する様子が画像化された。微小血管網における赤血球の分布状態に関しては、血漿層の形成状態、ファーレウス効果ならびに逆ファーレウス効果の様子が表示された。3.微小血管を流れる赤血球流速の低下にともない赤血球からの酸素放出の促進が観察された。pHが酸性になるにつれて、赤血球からの酸素の放出は増大した。4.血管の弾性が赤血球の微小循環動態(血漿層ならびに灌流抵抗)に及ぼす影響について、赤血球のレオロジー的性質を変化させて定量的に評価した。微小血管における赤血球からの酸素の放出への影響は今後の検討課題である。5.本装置の原理を応用して実体顕微鏡を用いて反射スペクトルによる解析を行い、脳表面の微小血管内を流れる赤血球の酸素飽和度の2次元画像を得ることができた。しかし、赤血球の流動挙動の定量的解析には、水浸対物レンズを装着した落射投光型顕微鏡を用いるのが適切であるとの結論に達した。
