新しい循環調節薬としてのエンドセリン変換酵素抑制薬開発のための研究

研究概要	エンドセリン(ET)は内皮細胞内でアミノ酸212残基の前駆体(ヒトの場合)より、アミノ酸38残基の中間体(bigET-1)を経て生合成される。bigET-1は血管収縮活性がほとんどない。したがってbigET-1からET-1への変換は生理的に重要であると考えられる。この変換を仲介するエンドセリン変換酵素(ECE)が実際に存在することは今までの研究からわかっていた。本研究計画ではこの変換酵素のcDNAを単離し、発現させ、その基礎的性質を調べ、変換酵素抑制薬開発の基礎とすることを目的とする。本年度はまずこのcDNA単離のための方法として、逆溶血反応法を開発した。ETを分泌していないCHOK1細胞にウシ内皮細胞cDNAライブラリーをトランスフェクトさせる。一方、ET-1の抗体を赤血球に結合させ、このET-1を分泌するCHO-K1細胞を溶血反応を指標として探索した。これによってウシ内皮細胞ET-1cDNAを分泌するCHO-K1細胞を単離し、最終的にECEcDNAを単離した。その結果ECEは蛋白部分の分子量約8万、膜貫通部位を1ケ所もち、亜鉛結合部位を持つ。中性エンドペプチターゼときわめて構造の似た金属プロテアーゼであることがわかった。さらにC末端側に10個のグリコシレーション部位を持つことから、C末端部を細胞外に持ち、したがつて活性部位を細胞外に持つと考えられる。さらにこのECEはbigET-1に特異的であるが、この他にET-3に特異的なECEも存在することがわかった。
