Gene Manipulationを用いたムンプスウイルスワクチンの作製

研究概要	新しいムンプスウイルスワクチンの作製するために、ネガティブストランドRNAウイルスにおけるリバースゲネティクスの確立をめざした。まず最初にパラインフルエンザウイルスやムンプスウイルスのReplicative complexの構成因子であるNP、P及びV蛋白の分子解剖を行った。V蛋白を発現させると核に局在することを見い出した。delation mutantを用いて核移行シグナルの同定を試みたところ、N末端(aa1-46)とC末端側(aa175-196)の2ケ所存在することが明らかになった。これらのシグナルは単独でも核移行に関与することが可能であるが、2ケ所含まれているほうが、核に留まる傾向が認められた。NP蛋白をV蛋白と共発現させたとき、単独に発現した場合には細胞質にのみ存在するNP蛋白が核内にも認められる。NP蛋白と結合して、核内に運んでいくに必要なV蛋白上の領域を解析したところ、核移行シグナルのうちのN末端(aa1-46)であることが明らかになった。P蛋白にはNP結合領域が2ケ所存在し、aa1-46とaa357-395であることを同定した。NP蛋白やP蛋白を単独で発現させると、細胞質内にdiffuseに存在するが、NP蛋白とP蛋白を共発現するとinclusion様に存在する。このようなinclusion様に存在するために必要なP蛋白上の領域を同定したところ、C末端側(aa357-395)であることが明らかになった。続いて、組替え体ワクチニアウイルスT7発現系を用いてParainfluenxa virusのリバースゲネティクスの検討を行った。現在full-lengthのcDNAの作製中である。またパラインフルエンザウイルスとムンプスウイルスとのキメラcDNAも作製中である。本研究によって、遺伝子工学に基づく新しいムンプスウイルスワクチンの開発に更に一歩近づいたと考えられる。
