研究概要	CTLエピトープの同定はきわめて複雑なため限られたものしか知られていない。そこで新しい考え方に基づいたリバース・イムノジェティックス法を用いて、HIV-1およびHTLV-1蛋白に対するCTLエピトープの同定を試みた。HLA-B^3501結合自己抗原ペプチドのモチーフ上のアンカー部位のアミノ酸をもった8-merから11-merのHIV-1およびHTLV-1ペプチドを合成し、HLA-B^3501分子との結合を調べた。HLA-B^3501細胞を用いたペプチド結合アッセイで、HIV-1ペプチドでは27種類のペプチドが、またHTLV-1では39種類のペプチドがHLA-B^3501分子と結合した。これらのペプチドを用いて、それぞれ2人のHIV-1感染者および3人のHAM患者の末梢血リンパ球を刺激し、細胞傷害性T細胞(CTL)を誘導した。HIV-1患者では、11種類のペプチドによってCTLが誘導することができ、そのうち10種類のペプチドで誘導したCTLがウイルスに感染した細胞を障害した。一方、HTLV-1患者では12種類のペプチドによってCTLが誘導でき、そのうち7種類のペプチドが誘導したCTLがウイルスに感染した細胞を障害した。以上のことから、リバース・イムノジェネティックス法によりHLA-B35分子によって提示することができるHIV-1およびHTLV-1CTLエピトープを効率的に多数同定することができた。今後他のHLAクラスI分子によって提示されるエピトープの同定をおこなう予定でいる。
