血管内皮・平滑筋細胞機能を細胞分子レベルで生体連続観察できるシステムの開発

研究概要	本年度は、血管内皮細胞や平滑筋細胞の〔Ca〕iや機能(EDRF産生、張力発生等)を、細胞・分子レベルで生体連続観察可能なシステムの開発に着手した。本システムでは、代謝要因(Ca Na,pH,NADH)の測定には指示蛍光色素および自然蛍光の2波長或は1波長蛍光測光を用いる。2次元蛍光情報の処理に使用するイメージプロセッサ(蛍光画像経時変化測定システム)は、フルサワラボより購入し、必要に応じて改良を加えた。基礎実験で、現有の光学系(日本分光:CAM230)の光量はこのイメージプロセッサのシグナルとして不足とわかったので、旭計器工業に新しい光学系の作製を依頼した。これらの変更によって、比較的遅いシグナル(秒単位)は計測可能となったが、これより速いシグナルは未だに計測出来ない。この件に関するプログラムを現在開発中である。本システムを利用し、血液凝固因子トロンビンの血管内皮および平滑筋細胞に対する作用を検討した。豚の(1)大動脈弁内皮細胞、(2)冠動脈中膜平滑筋細胞、(3)内皮付冠動脈中膜平滑筋細胞の〔Ca〕iを、フラ-2蛍光測光した。(1)内皮細胞の〔Ca〕iはトロンビンによって一過性、濃度依存性に上昇した。(2)U46619によって前収縮させた中膜平滑筋はトロンビンにより、更なる収縮と〔Ca〕i上昇をしめした。(3)内皮付平滑筋は、i)脱分極収縮の場合、トロンビンによって弛緩するが、これはnitro-L-arginineで完全に抑制されるため、EDRFが関与していると考えられる。ii)U46619収縮の場合、トロンビンは一過性、濃度依存性に〔Ca〕i低下と弛緩を引き起こすが、nitro-L-arginineはこの弛緩を部分的にしか抑制出来なかった。これは、EDRFのみならず、EDHFもトロンビンの弛緩作用に関与することを示す。トロンビンはこのように収縮・弛緩の両面から血管の緊張を制御している事が明らかとなった。
