遺伝子工学的手法を用いた新しいヒト脳腫瘍マーカーの単離と解析

研究概要	我々は、ラット脳の発達過程で胎生期には豊富に発現されるが成熟期の脳では発現が減少する胎生期脳選択的mRNAのcDNAクローンをこれまでに22種類単離している。本年度は、ヒトの相同クローンを単離して、ヒト脳腫瘍におけるmRNA発現の解析を行なった。また、ヒトグリオーマ細胞株を用いて、腫瘍細胞の悪性度に関与する可能性のある遺伝子群の単離と解析を行なった。ヒトのEST(expressed sequence tag)データベースを有効に利用しながら胎生期脳選択的クローンのヒト相同クローンを単離し、Northern blot法でヒトでの発現パターンを解析した結果の中では、Neuronatinの発現パターンが特に注目された。即ち、ヒトNeuronatin mRNAは胎生期の脳で選択的に強く発現されており、成人の多くの組織では発現を認めなかった。また、Neuronatin mRNAは解析したヒト各種の脳腫瘍の中で、下垂体腺腫に選択的な発現を示し、ホルモンを産生しない非機能性下垂体腺腫でも強い発現が認められた。一方、増殖率と異種移植能の異なる2種のヒト培養グリオーマ株におけるmRNA発現パターンの違いを解析した結果、増殖速度が早くヌードマウスへの異種移植能を持つグリオーマ細胞株では、蛋白合成、シグナル伝達、核酸代謝等に関わる様々な遺伝子の発現量が対照株に比べて増加していることが判明した。特にIL-8の発現量が1,000倍以上に増加していることが注目された。これらの知見は、近い将来、新しいヒト脳腫瘍マーカーの同定につながりうる重要な所見であると考えられる。
