研究概要

我々は、NIH3T3細胞のv-srcトランスフォーマントの細胞破砕液を用いた、複数のプロテインキナーゼ活性を同時に検出できる系を開発し、細胞内スグナル伝達解析への応用を試みてきた。このアッセイ系は、プロテインキナーゼ阻害剤の探索系として適しており、選択性の高いプロテインキナーゼ阻害剤開発を目標に、今年度、この系を用いたスクル-ニングを開発した。また我々はこれまで、癌細胞の性質を正常様にする物質の探索やその作用機構の解析を行ない、シグナル伝達研究に役立ててきた。今年度新たに、培養系における癌細胞の性質の中で、造腫瘍性と最も良く相関するといわれる足場非依存性増殖を、親水性ポリマーでコートした96穴プレート中で測定できる方法を開発した(印刷中)。癌細胞に特有の性質である足場非依存性増殖を選択的に抑制する物質は、癌化シグナル抑制物質であることが予想されるが、従来の軟寒天コロニー形成法は操作が煩雑であり、定量性に乏しく、時間も費用もかかることから、スクリーニングに適用することは事実上不可能であった。我々の開発した方法は簡便、迅速、安価でかつ定量性であり、スクリーニング系として優れた点が多い。予備実験を行ない、この系で足場非依存性増殖抑制物質を探索できる可能性が示されれば、スクリーニングを開始する。またこの方法では足場非依存的に増殖している細胞の回収が容易であるため、従来法では困難であった生化学的解析が可能である。上記のキナーゼ活性測定法と組み合わせ、足場依存性の正常細胞と足場非依存性の癌細胞のシグナル伝達の違いを調べる予定である。
