トランスゲニックマウスを用いた多能性幹細胞の不死化と分化誘導

研究概要	多能性造血幹細胞を不死化し分化誘導させるために、まず、T-抗原トランスジェニックマウスから多数の骨髄間質細胞株を樹立し、セルソーターを用いて純化した幹細胞の増殖・分化の制御を解析した。その結果、2つの特徴的な幹細胞分化誘導能をもつ間質細胞株を発見した。1つ(TBR59)は幹細胞のミエロイド、リンパ球両系への分化を誘導し、他(TBR31-1)はリンパ球系への分化のみを誘導した。両間質細胞は直接の細胞間相互作用により幹細胞を制御しており、c-Kit,SCFあるいはVCAM-1,VLA-4の分子間相互作用が必須であることが分かった。この2つの間質細胞株は、前脂肪細胞株であり、一方は主として幹細胞をミエロイド系へ分化誘導し、他はリンパ球へと分化誘導することが分かったので、両者にそれぞれ特異的に発現している細胞表面タンパクのモノクローナル抗体によるスクリーニングを行い、いくつかの特異的抗体を産生するミエローマクローンを得ることができた。また、TBR31-1株との共培養からBリンパ球の極めて幼若な幹細胞株が樹立され、表面抗原のセルソーターによる解析とRT-PCRによる遺伝子発現の解析からリンパ球系幹細胞であると結論した。また、他の間質細胞株とサイトカインの組み合わせにより、段階的に成熟B細胞まで誘導する系が構築できた。これらの結果から、血液幹細胞の分化方向の決定が間質細胞より制御されていることが明らかとなった。さらに多能性幹細胞を不死化するために、p53ノックアウトマウスからソーティングした造血幹細胞を間質細胞株上で共培養すると、より長期間維持できることが分かり、不死化への基礎的検討ができた。
