研究概要

ムスカリン受容体m2サブタイプの変異体を定常的に発現させる系を確立した。この変異体は糖鎖結合Aspを変異させ、第三細胞内中央部を除去し、C末端にヒスチジン6残基を融合したもので、発現効率がよく、簡便に精製できることがこれまでの研究で分かっている。約50nmolの受容体を含む21のSf9細胞培養液が1週間で得られる。200nmolの受容体を含む膜標品からCoカラムを用いた単一操作で約50nmolの部分精製標品の調製が可能である。受容体の可溶化、精製にはジギトニンを使用しているが、ジギトニンはリン脂質平面膜の生成を抑制するという問題点がある。種々の界面活性剤の検討を行い、テトラデシルマルトシドにより受容体が比較的安定なことを見出した。現在、ジギトニンをCHAPSまたはテトラデシルマルトシドに変えた標品について2次元結晶化の試みを行っている。受容体は並ぶ傾向が見られるが、結晶化には至っていない。
受容体に結合したリガンド分子のコンフォメーションを解析する目的で、メタコリン(O-acetyl-β-metylcholine)の光学異性体を乳酸から合成した。光学異性体s-メタコリンと部分精製m2受容体の混合物でTRNOEの測定を行っている。非特異的結合に由来する信号の分離を検討中である。
