研究概要

1.ハンタウイルス組み換え核蛋白をバキュロウイルスを発現ベクターとし、カイコ虫体内に発現させた。不溶性画分に主に存在する発現蛋白を、初年度までに開発した低速遠心法で分離・精製したあと、各研究分担者に供給し、ELISAとWestern blotting法による感度と特性をIFA法と比較・検討した。
2.バキュロウイルス発現ハンタウイルス組み換え核蛋白(Bac.HTN-NP)を用いたELISAでは、ラット、ヒト血清いずれにおいても感度はIFA法と同等であった。しかし、陰性血清におけるバックグランドの発色のレベルが比較的高く、発色の程度を肉眼判定するには問題点が残った。
3.上記2.で明らかとなったバックグランド発色は、抗原の精製の不十分さが原因と考えられた。しかし、Bac.HTN-NPは、ELISA抗原として使用するための穏やかな条件下では可溶化されづらくこれ以上の精製が困難であった。そこで、次に、大腸菌中に同様のHTN-NPを発現させる系を構築した。この系では発現蛋白にビオチンが融合蛋白として結合しているためにアビジンを用いてcapture ELISAを確立した。現在、本法での感度・特異性を検討中である。
4.Bac.HTN-NPをWestern blotting法に応用した場合、感度はラット血清、ヒト血清いずれにおいてもIFA法とほぼ同等であった。しかし、陰性血清群を用いた特異性の検討では、IFA法で非特異反応や紛らわしい蛍光像を示す検体でもWestern blotting法では非特異反応が全く認められなかった。すなわち、Western blotting法は、IFA法に比べ特異性が高いことが判明し、確定診断法として有用であると考えられた。
