研究概要

◆本年度9月に、まず東京大学臨海実験所新実験研究棟の飼育実験室内に、海水濾過装置および給餌装置を装備した海産動物飼育培養装置(アクアKK・CH-50)を製作し、設置したので従来より軽作業で純系海産生物の飼育培養が可能となり、本研究遂行に大いに働いている。本装置を使用し、1985年に野生株より分離した固体をもとに開始したカタユ-レイボヤ自家交配系は、17代目までに達した。一時期、生殖・成長能力の劣えが見られ存続が危ぶまれたが、最近1年間は安定した生殖・成長能力を示している。この自家交配系(純系)の特性を検定するために、移植拒絶を指標とした免疫学的検定とDNA塩基分析による検定を準備している。◆岡山大学臨海実験所では、ヒトデ純系作出の基礎として、受精卵から生体までの飼育方法の確立を試みている。浮遊珪藻で飼育した幼生は3週間で変態、その後付着珪藻を与えて飼育した。70日後の今、径2mmまで成長、幼体に適した餌を探索中である。◆アスキジア科のホヤは、バナジウムを高濃度に濃縮する特異な生理機能を有する。そのため生体の金属イオンの濃縮機構を研究するには格好のモデル動物である。広島大学臨海実験所では、アスキジア科のナツメボヤとスジキレボヤを、シオナ科のカタユレイボヤをそれぞれ選んで屋内外での養殖を試みた。その結果、本年度は屋内水槽で3代までの継代飼育を行うことができた。◆京都大学では、ナメクジウオ,ホヤ,ギボシムシのゲノムライブラリーの作成を試み、ナメクジウオおよびホヤでは完成した。現在ジボシムシのライブラリーを作成している。◆東京工業大学では、ユウレイボヤの純系を作成するための前段階として全国各地よりホヤを採集し、rRNA遺伝子スペーサー領域を指標としてPCRを用いて多系を調べた結果、採集地点および個体により変異があることが見い出された。今回作成されつつある純系についてこの多系について検討している。
