突然変異体を利用したイノシトールミリン酸受容体の機能解析

研究概要

味覚トランスダクション機構にどのようなセカンドメッセンジャーに関わっているかはまだ解明されていない。これまでのショウジョウバエを用いた研究によって、norpA視覚突然変異体において味覚異常がみられることから、イノシトール3リン酸(IP_3)がセカンドメッセンジャーとして機能していることを明らかにした。ショウジョウバエのIP_3受容体遺伝子は感覚器において高い発現があることから、トランスダクション機構にIP_3受容体が関与していることが示唆される。本研究では、IP_3受容体の突然変異体を分離することによって、IP_3の味覚受容における役割を解明することが目的であった。IP_3受容体遺伝子が位置する座位にはこれまで分離されている突然変異体は報告されていない。近傍にP因子が挿入された複数の系統についてサザン解析を行ったが、IP_3受容体遺伝子内へのP因子の挿入は確認できなかった。そこで、IP_3受容体遺伝子座の近傍にP因子が挿入された3系統を用いて、local jump法により多数のP因子挿入致死系統を分離した。また同時に、imprecise exicisionによる欠失系統を分離した。これらの系統がIP_3受容体遺伝子内にP因子の挿入あるいは欠失があることを確認している。
