組換え体セントロメア蛋白質のDNA結合能を用いたセントロメアDNAの単離

研究概要

1。DNA結合能を持つ新たなセントロメア蛋白質をコードする遺伝子の同定
DNA結合活性を持つセントロメア蛋白質(CENPs)を同定するため、ヒトcDNAライブラリーより抗セントロメア血清に反応するクローンをスクリーニングし、この中からセントロメア蛋白質B(CENP-B)、セントロメア蛋白質C(CENP-C)、ヘテロクロマチン蛋白質1(HP1)をコードする遺伝子を得た。大腸菌に発現させた組換え体蛋白質を用いた解析により、これらがDNA結合能をもつことを明らかにした。続いて、ゲル移動度シフト法、サウスウェスタンブロット法によりこれらの蛋白質のうち、DNA結合能を持つ領域の限定を行った。その結果、CENP-BについてはN末端135アミノ酸残基の領域に、CENP-Cは内部の104アミノ酸残基の領域に、HP1については内部の64残基の領域に存在することが明らかとなった。また、CENP-CとHP1については、そのアミノ酸配列に類似性が存在した。
2。DNAアフィニティー沈降法を用いたCENP-B結合配列の単離
ヒトCENP-Bについてはセントロメア領域のサテライト(アルホイド)に存在する幾つかの配列に結合することが知られているが、その結合配列の詳細は不明であった。そこで、標的配列を単離・解析するため、ヒト組換え体CENP-Bを用いたDNAアブィニティー沈降法を開発した。これを用い、ヒトゲノムライブラリーより、CENP-Bの結合するセントロメアルホイドDNAを単離した。実際、塩基配列を解析した結果、新たなCENP-B結合配列が存在していた。
