研究概要	本研究はレニン産生細胞とターゲットとしてより高感度なハイブリッド組織化学法を開発することにある。本年度はまず、代表的なレニン産生臓器である腎臓を用いて本法を行い有効なプローブ濃度、proteinase Kの反応条件、ハイブリダイゼーション温度の設定を検討した。その結果、proteinase Kは0.25mg/ml,10分の反応条件で十分目的が達せられ、これ以上の反応条件ではむしろ形態の破壊や切片の剥離などの悪影響をもたらすことがわかった。またハイブリダイゼーション温度は55℃で最適な結果が得られた。本法によって腎臓の糸球体傍細胞に強いシグナルの局在が認められた。次に、従来より検討してきた凝固腺レニンの検出を同法に従って行った。コントロールではその上皮細胞内に明瞭なシグナルの局在が観察されたが、去勢後4週では認められなかった。Reverse transcriptase-Polymerase chain reaction(RT-PCR)法を併用しても、この例ではバンドは検出されなかった。次年度はRT-PCR法の組織応用により最適な臓器の選択から検討する必要性がある。
