神経再生機構の加齢による影響について

研究概要

末梢神経損傷後、再生軸索が骨格筋に進入し、脱神経された神経筋接合部(neuromuscular junction.NMJ)を再支配する機構が老化によりいかなる影響を受けるかについて解析した。若年期(3月令)に比較すると老齢期(24月令)ラットでは次の特徴がみられた。1.再生軸索の筋内進入時期は遅れる。2.神経線維束内にて増殖する大食細胞はより長い期間存在する。3.軸索、細胞外物質・レクチン(PNA)、NMJ(bunngarotoxin陽性部位)の三者の相互関係を蛍光組織化学的に重染色法により解析した。再生軸索は(1)屈曲蛇行、(2)NMJへの方向性の喪失を示し、NMJのPNA陽性部位は顕著な分布異常(変性物質の出現、不均一な分布)を示した。これらの所見は再生軸索のシナプス認識機構における異常を反映する。現在の研究展開。1(in vivo).神経損傷部位、脊髄前角における再生活性の発現様式。2(in vitro).単離された毛様体神経節細胞と脱神経筋よりえられたNMJ試料をco-cultureし、神経細胞からの突起とNMJ部位の再結合、相互関係を解析する。
