研究概要

(A)アフリカツメガエル卵細胞に発現させた血管作動性腸管収縮ペプチド(vasoactive intestinal contractor:VIC)の研究
1)VIC受容体は、ラット腸管由来のmRNAをアフリカツメガエル卵細胞に注入する事によって発現させた。
2)双電極膜電位固定法(two-electrode voltage-clamping method:保持電位-60mV)下で卵細胞に低濃度(1nM〜100nM)のVICを投与すると、濃度依存性に内向き電流が誘発された。
3)VIC受容体の数は、日数と共に増加した。
4)VIC反応は、Ca^<2+>依存性Cl^-チャンネルの開口によるCl^-電流である。
5)VICを卵細胞に反復投与すると、内向き電流反応は次第に小さくなると共に時間経過が緩やかになるという脱感作(desensitization)現象が見られた。
6)アミノ酸構造が酷似している三種のエンドセリン(ET-1,ET-2,ET-3)のいずれにも反応しないのにVICのみに反応する場合があった。即ち、VIC受容体はエンドセリン受容体とは異なるものであり、「VICはエンドセリン受容体に働いて作用を及ぼすに過ぎない」という現在の主要な説を見直す必要がある。
卵細胞に発現させたVIC受容体の性質を詳しく解析する為に、卵細胞内でのCa^<2+>遊離機構を調べる必要があったので、以下の実験を行った。
(B)アフリカツメガエル卵細胞内の遊離機構の研究
細胞外液のpHをアルカリ性にすると、細胞内でのCa^<2+>遊離を抑制すると共に卵細胞膜に存在するK+チャンネルを開かせて外向き電流反応を生じた。この電流は、細胞内のCa^<2+>濃度とは関係がなく、cyclicAMP系を介して活性化されるものであった。一方、細胞外液のpHを酸性にすると、細胞内のCa^<2+>貯蔵所からCa^<2+>が遊離されてCa^<2+>依存性Cl^-チャンネルが開き、内向き電流反応を生じた。
卵細胞周囲の体液のpHを測定すると8に近いアルカリ性であり、これが排卵前の卵細胞内でのCa^<2+>遊離を抑制して卵細胞の成熟を抑制しているという生理的な意義を持っていると考えられた。
