脳内プロスタグランジンによる抗利尿ホルモン分泌促進機構の解明

研究概要

1.ラットの静脈内に高張NaCl溶液を注入すると、15分、30分後に、血漿浸透圧、Na^+、Cl^-の増加を抗利尿ホルモン(ADH)濃度の上昇が惹起された。このADH反応は、NaCl注入の30分前にPG産生阻害薬メクロフェナメイト(MCL)を第三脳室腹側前部(AV3V)に局所微量投与すると、完全に消失した。MCLはADH以外の、血漿浸透圧、電解質、血圧等の因子の反応には影響しなかった。これらのラットにおけるMCLの投与部位は、終板脈絡器官やその近傍、視索前野正中核、内側視索前野、脳室周囲核に認められた。これらの部位にPGE2を局所投与すると、5分および15分後、血漿ADHと動脈圧が著しく増大した。血漿浸透圧や電解質の濃度は変わらなかった。薬物投与用のカニューレが以上の部位をはずれて脳室内に入った場合や、AV3Vに投与したものと同じ量を故意に脳室内に与えた場合には、MCLやPGE2投与の効果は認められなかった。従って、投与した薬物が、脳脊髄液流に乗って運ばれ、AV3V以外の領域で働いたとは考え難い。他方、MCLのAV3V内投与は、等張NaCl溶液の静脈内注入下にみられるADH、その他の因子の反応には影響しなかった。
2.ラジオイムノアッセイによるPGE2の測定法を確立し、PGE2産生におよぼす浸透圧増加の影響を調べた。ラットの静脈内に等張又は高張NaCl溶液を注入し、経時的に断頭採血した。血漿PGE2濃度は、どちらの溶液の注入によっても、有意に変化しなかった。しかし、実験前日にAV3Vに植え込んだ微小透析プローブを介して採取される間質液中のPGE2濃度は、60分間にわたる高張NaCl注入の最初の30分間に、増大する傾向を示した。
以上の結果は、AV3Vあるいはその近傍で産生されるPGは、高浸透圧状態におけるADH分泌の高進に寄与することを示唆している。
