神経内分泌ニューロンにおける性ホルモンによる可塑性の発現

研究概要	我々はこれまで、視床下部のオキシトシン分泌ニューロンにオキシトシンを投与すると、雄ラットではニューロンは促進反応を示すが、virgin雌では抑制反応を示し、第一仔の分娩時に促進反応に逆転し、授乳が終了すると元の抑制反応に戻る事を明らかにして新しいタイプのニューロンの可塑性を見出した。本研究ではこの可塑性を発現させる因子の究明を第一の目的とし、以下の結果を得た。(1)卵巣切除したラットの視索上核を薄切片として摘出し、微分干渉顕微鏡下で細胞外記録を行い、ニューロンのオキシトシンに対する反応を調べたところ促進反応を示した。よって、ニューロンのオキシトシンに対する抑制反応発現には卵巣から分泌される因子が必要である事が解った。(2)卵巣切除したラットにエストロゲンを投与するとオキシトシンは抑制反応を惹起する事から、反応逆転を引き起こす因子の少なくとも一つはエストロゲンである事が解明した。(3)エストロゲンを投与する直前にアニソマイシンを投与し、蛋白質の合成を阻害するオキシトシンはニューロン活動を促進したので、エストロゲンの作用発現には新たな蛋白質の合成が必要である事が解った。(4)インタクトのvirgin雌ラットをアンチ・エストロゲンであるタモキシフェンで処理すると促進反応が出現するので、エストラゲンが視床下部のニューロンに作用し可塑性を引き起こす因子である事が判明した。(5)上記の実験における抑制反応、促進反応はどちらも同じオキシトシン・リセプタ・アンタゴニストで反応が阻止できる事から、同一レセプタを介する反応と考えられる。
