研究概要

我々は、哺乳類の神経分化機構を転写制御因子のレベルで明らかにする目的で、ショウジョウバエの神経分化に重要な役割を果たすhairyというHLH型因子の哺乳類ホモローグHES-1について解析をおこなってきた。その結果、HES-1は神経分化因子Mash-1をターゲットにする分化抑制因子であり、その発現低下が中枢神経分化の進行に重要であることを明らかにした。本研究では、HES-1の標的破壊マウスを作製して更に詳細なHES-1の機能解析をおこなうとともに、新たな神経分化因子の同定を試みた。
HES-1の標的破壊マウスは脳胞形成に異常がおこり、胎生致死か出生直後に死んでいた。従って、HES-1は神経分化抑制作用をもつが、神経の初期発生において必須の因子であることが明らかになった。
次に、新たな神経分化因子を同定するためにPCR法により検索したところ、新しいHLH型因子をコードする2種類の遺伝子を得た。この因子はいずれもショウジョウバエの神経分化因子atonalによく似た構造をしているのでMATH-1およびMATH-2と名付けた。いずれの因子も発生途中の神経系に特異的に高レベルに発現していた。さらにトランスフェクションの実験から、MATH-1はMash-1と同様に転写活性化作用をもち、その活性はHES-1により完全に抑制された。以上の結果から、この新しい因子はHES-1により抑制される正の神経分化制御因子の有力な候補となることが示唆された。
哺乳動物の神経分化においても、ショウジョウバエと同様、HLH型因子が重要な役割を果たしていることが明らかになってきたが、まだ関与する因子の一部が同定されたにすぎない。今回我々が新たに同定した因子についても遺伝子標的破壊マウスを作製することにより神経分化機構がより一層明らかになるものと思われる。
