ホルモン依存性プログラム細胞死の開発と応用

研究概要	1研究目的本研究は、プログラム細胞死を脂溶性ホルモンにより自由にコントロールしうるシステムを作り出すこと,このシステムを応用発展させることにより、全く新しい癌の遺伝子療法を確立することを目的としている。さらにこのシステムと動物への遺伝子導入法を組み合わせることにより、種々の変性疾患モデル動物の作製を試みる。 2研究成果 fas抗原受容体のプログラム細胞死に必須な蛋白領域とエストロジェンホルモン受容体もしくはレチノイン酸受容体のリガンド結合領域を結合した融合蛋白質をコードする融合遺伝子を作出し、fas抗原に感受性があることが知られているL929細胞及びHeLa細胞に恒常的に発現するように導入した。得られた細胞株に、導入した融合遺伝子の受容体部分に対応するリガンド(エストラヂオール、レチノイン酸)を加えたところ、どちらの細胞においても0.1nMのエストラヂオール、10nMのレチノイン酸で処理後約3時間ですべての細胞が死滅することが観察された。つまり、現時点で少なくとも培養細胞レベルにおいて脂溶性ホルモンによりプログラム細胞死を自由にコントロールしうるシステムの確立に成功した。現在、遺伝子導入したL929細胞及びHeLa細胞をヌードマウスに移植しin vivoにおいても細胞死が脂溶性ホルモンにより誘導されることを確認する実験を行っている。また、このシステムに感受性のあるヒト癌細胞株を系統的にスクリーニングしているところであり、これにより将来的に治療の標的となりうる癌の種類が特定できると考えている。さらに、in vivoの癌細胞に効率よく融合遺伝子を導入するために、レトロウィルス、アデノウィルス、リポゾームを用いる方法を検討している。また、これらの融合遺伝子を小脳特異的なプロモーターの支配下に繋いだ遺伝子を導入したトランスジェニック・マウスを作りつつある。このマウスに脂溶性ホルモンを与えることにより、小脳変性疾患のモデルマウスの作出を試みる。
