大腸発癌背景粘膜上皮に於けるapoptosis,サイトカイン発現の変動の解析

研究概要

今年度は、大腸発癌背景粘膜と大腸腫瘍におけるサイトカインを特にtumor necrosis factor-α(TNF-α)とinterleukin-1α(IL-1α)について検討した。
[方法と材料]F344雄ラットを用い、化学発癌物質methylazoxymethanol(MAM)acetateと1-hydroxyanthraquinone(1-HA)投与を行い、大腸腫瘍と炎症性病変を作成した。特に1-HAは潰瘍性大腸炎様の病変を誘導し、サイトカインの発現とその変動を検討するのに適当であった。TNF-αとIL-1αの発現は病変からのRNA抽出によりreverse transcriptase-polymerase chain reaction(RT-PCR)法によりそれぞれのmRNA発現を検討した。
[結果・考察]1-HA投与を行った2群(MAM acetate+1-HAと1-HA単独)の非腫瘍性大腸粘膜において、TNF-αとIL-1αの発現はともに対照群に比し増加しており、1-HAが誘導する炎症性背景による影響と考えられた。MAM acetae単独群での非腫瘍性粘膜においても対照群に比し増加傾向を認めた。一方、すべての誘導した大腸腫瘍においては、上記の非腫瘍性粘膜での発現に比べ、より増加傾向を示し、TNF-αとIL-α等のサイトカインの腫瘍形成への関わりが示唆された。
こうしたサイトカインの発現とアポトーシスとの関連性および局在について、次年度検討していく予定である。
