研究概要	研究代表者はEDINのVero細胞に及ぼす影響について検討し、EDINがVero細胞のゴルジ体を解離させることを見出している。この作用はbrefeldinAとよばれる細胞内膜系に影響を与える抗生物質の作用と酷似しているが、rho p21がどのような形でのこ作用に関係しているのかは明らかにされていない。研究代表者はVero細胞を用いてEDINの基質について検討し、Vero細胞が等電点の異なる複数の基質を有することを明らかにした。またこの基質は、細胞膜、細胞質の両方に存在することが明らかになった。またbrefeldinA処理あるいはEDIN処理によって細胞膜画分のEDIN基質は蔗糖密度勾配遠心分画法によって比重の大きなゴルジ体領域から比重の軽い方へ移動することが明らかとなった。また、この研究を行っている過程で病原性大腸菌が産出するCytotoxic Necrotizing Factor 2(CNF2)がEDINの標的であるrho p21を修飾することを見出した。このCNF2によるrho p21の修飾によってrho p21の見かけ上の分子量が大きくなる。この修飾の本体は不明であるが、CNF2の作用によって細胞は見かけ上、rho p21の機能を活性化された性状をとる。CNF2の遺伝子cnf2とrhoAを共発現させた大腸菌内で、発現したrhoA p21が修飾されたことからCNF2が酵素作用有している可能性について現在検討中である。
