研究概要	全世界でコレラ菌のコレラ毒素(CT)、毒素原性大腸菌の易熱性エンテロトキシン(LT)が原.因の下痢症で約150万人の小児が死亡し、ワクチン開発が急務とされる。一方両毒素はアジバント活性を持ち、有効な変異毒素の作製が望まれている。そこで我々はAサブユニット(A)の変異毒素を作製し、毒素活性増殖因子(ADPリボシル化因子)のCTへの作用部位を解明することにより、無毒で臨床的に有効な変異毒素の作製に寄与することを目的に本研究を企画した。我々はAの変異毒素(E112K)を用いて、ARF(ADPリボシル化因子)の毒素への作用様式がアロステリック効果によると明かにしている。そこでGlu112を含めて近傍及び遠隔アミノ酸を色々置換した変異毒素を作製する事により、その影響範囲を決定を試みた。Glu1122近傍の活性洞を形成するアミノ酸を遺伝子工学で目的のアミノ酸に変換した変異毒素作製し、既に94年度に作製した酵母菌からのARF遺伝子を用いて作製したrARFとの相互作用を検討した。その結果Glu112を中心に形成されている活性洞に直接ARFが結合して毒素の酵素活性を上昇させるのでは無く、一個のアミノ酸を通じて活性が上昇する事を明らかにした。さらに標的蛋白質及びpolyarginineのADPリボシル化に関与する、すなわち毒素の標的物質に結合する部位の固定に成功している。これらの結果は毒素が如何にして標的蛋白質を選択しているかを解析できる基礎となる。
