化学合成によるエンドトキシンアンタゴニスト構造の解明とその応用

研究概要	細菌感染に伴うエンドトキシン疾患の臨床上の重要性から、エンドトキシンアンタゴニストの開発への期待は大きい。ここでは、無毒性でアンタゴニスト活性を示すリピドA類縁構造を化学合成手法を用いて解明し、その応用への発展を図ろうとするものである。著者は天然のリピドAをサクシニル化することできわめて効率的に強力なエンドトキシンアンタゴニストを誘導することができることを見いだした。このアンタゴニスト作用はエンドトキシンの示すマウスB細胞マイトジェン活性、マクファージの活性化(TNF,及びNO産生)さらには人由来THP-1細胞のTNF産生いずれに対しても見られた。化学合成リピドA類縁体を用いた解析より、E.coliやSalmonella等の完全構造リピドAは、この修飾によってエンドトキシン活性には全く影響を受けなかったが、前駆体構造を同修飾すると完全に活性がなくなり、強力なアンタゴニストに変換することを見いだした。この結果は還元末端の遊離水酸基は活性に全く影響を与えないが、非還元末端側の脂肪酸の遊離水酸基の置換基はエンドトキシン活性、アンタゴニスト構造の誘導に決定的な役割を果たすことを示唆する。このことを直接証明すると共に、この前駆体構造の遊離水酸基のうち、どれが活性構造、アンタゴニスト構造を決定しているのかを確定するために、特定の水酸基のみにサクシニル基を導入した化合物群の合成を行った。その結果、3'位脂肪酸の3位水酸基をサクシニル基で修飾するだけで前駆体が完全にエンドトキシン活性を喪失していること、さらには強力なアンタゴニスト作用を示すことが明らかとなり、この部位の水酸基の置換基が活性発現、アンタゴニスト構造誘導に重要な働きをしていることが証明された。現在、それ以外の遊離水酸基にそれぞれ一個のサクシニル基を導入した化合物群を化学合成途上にあり、その解析は極めて重要な情報を与えるものと期待される。
