ヒト免疫グロブリンH鎖遺伝子全領域の構造解析

研究概要	1)新たに単離した2個のYACクローンのうち、11Iは既存の0.8Mbの領域の最上流からさらに約50kb上流に伸びたクローンあり、未知の領域には新たな3個のVH断片(V3-65P、V3-66、V1-67P)が同定された。一方クローン13.3は全長約140kbであり、クローン11Iを含む下流の領域とは異なる領域由来であった。このクローンは12個のVH断片を含んでおり、また興味深いことに、領域の最上流にヒトテロメア特異的反復配列が見い出された。クローン11Iと13.3の間に残るギャップの大きさを明らかにするため、各々の領域のコスミドクローンを用いた間期染色体のFISH法を行なった。その結果、2つの領域の間は約110kb離れていることが明らかになった。以上を総合して、染色体第14番上のヒトVH領域の全長は約1.1Mbであることが明らかになった。 2)ヒトゲノム中に存在するVH断片のおおよその数が、今回の解析より明らかになった。我々が現在までに単離を終了した領域には81個のVH断片のうちV1-68、V1-69を除く合計79個のVH断片を有し、また個体により有無に差のあるVH断片が現在までに6個(V7-4.1、V4-30.1〜V4-30.5)報告されており、さらに未知のVH断片が残されたギャップ中に4〜5個程度存在すると推測している。以上から、VH断片の総数は個体による差があるが、81個+10〜11個程度であろうと結論した。これに加えて、染色体第15番、16番上にそれぞれ7〜11個、14〜17個のVH断片が存在することが判っている。以上を総合して、ヒトゲノム中のVH断片の総数は102個から120個の間であろうと結論した。
