研究概要

インターロイキン2受容体(IL-2R)を介する増殖性シグナル伝達において、Jak3チロシンキナーゼ(PTK)が必須の役割を担うことが明らかとなった。Jak3 PTKは、IL-2刺激に伴うc-myc及びc-fos遺伝子の発現誘導に重要な役割を担うことも明らかとなった。また、今回新たにIL-2シグナルの標的遺伝子としてチロシンフォスファターゼLC-PTPを同定することが出来た。IL-2Rβ鎖の構造と機能の連関解析により、IL-2Rβ鎖の細胞内″serine-rich″、 ″acidic″両領域がLC-PTPの発現誘導に必須であること、加えてIL-2Rβ鎖の細胞内C末端側半分の″H″領域がSTAT (Signal Transducer and Activator of Transcription)5の活性化に重要であることが明らかになった。STAT5の活性化に重要なIL-2Rβ鎖の″H″領域は、IL-2刺激に伴う細胞増殖には必須でないことから、STAT5は細胞増殖以外の細胞応答において重要な役割を担っている可能性が示唆される。さらに、キナーゼの自己リン酸化能を利用した″in gelキナーゼ解析法″により、IL-2Rβ鎖の細胞内領域と選択的に会合する新規セリン・スレオニンキナーゼ(分子量約80、85及び90kDa)を同定することが出来た。これら3つの異なる分子量を有するセリン・スレオニンキナーゼは、フォスフォペプチドマッピング解析から同一のキナーゼ分子が異なる修飾を受けたもの、あるいは類似のキナーゼであることが示された。
