イヌ小腸単離上皮細胞を用いたモチリン分泌刺激因子に関する研究

研究概要

1.消化管単離上皮細胞を用いた、細胞潅流実験系の確立
コラゲナーゼを用いて単離した細胞を、BSAを含む生理的食塩水にて潅流した。潅流液中のモチリン濃度はRIA法にて測定し、刺激に対して再現性の良いモチリン分泌が見られた。モチリン分泌は、二つのピークより成り、高濃度のアセチルコリン付加により、二つ目のピークが顕著に現れた。この反応は、下垂体単離細胞よりの下垂体ホルモン分泌を潅流実験系を用いて観察するときに見られる反応と同様の物と考えられる。
2.アセチルコリンに対する反応
アセチルコリン投与により、10^<-4>Mにピークを持つ容量依存性のモチリン分泌反応が得られた。この反応は、アトピン同時投与で容量依存性に消失することより、ムスカリニックレセプターを介していると考えられる。
3.ソマトスタチン、ボンベシンによる分泌反応
今までにモチリンを分泌を調節するとin vivo実験にて報告されていたソマトスタチン、ボンベシンは、モチリン分泌を変化させなかった。従って、これらのホルモンによるモチリン分泌調節はモチリン産生細胞への直接作用とは考えられない。
4.アセチルコリン神経繊維の免疫染色
アセチルコリンに対する抗血清を用いて、小腸絨毛内のアセチルコリン神経繊維を免疫染色した。アセチルコリン免疫陽性繊維は、小腸上皮細胞の直下に見られた。
今後は、アセチルコリンレセプターサブタイプの確認と、モチリン細胞上の証明を中心に研究を進めたい。
