イヌ小腸単離上皮細胞を用いたモチリン分泌刺激因子に関する研究

研究概要	1.消化管単離上皮細胞を用いた、細胞潅流実験系の確立コラゲナーゼを用いて単離した細胞を、BSAを含む生理的食塩水にて潅流した。潅流液中のモチリン濃度はRIA法にて測定し、刺激に対して再現性の良いモチリン分泌が見られた。このモチリン分泌は、二つのピークより成り、高濃度のアセチルコリン刺激により、二つ目のピークが顕著に現れた。この分泌様式は向下垂体因子による下垂体前葉細胞からのホルモン分泌と一致しており、、消化管ホルモン分泌の潅流実験系が確立できた。 2.カルバコールに対する反応とムスカリニックレセプターのサブタイプの決定カルバコール投与により、10^<-4>Mにピークを持つ容量依存性のモチリン分泌反応が得られた。この反応は、アトピン同時投与により10^<-9>Mより有意に抑制され、10^<-5>Mで完全に消失した。ムスカリニックレセプターのサブタイプを調べるため、M1, M2, M3のアンタゴニストとしてそれぞれピレンゼピン、AF-DX116BS、4-DAMPを用いて、カルバコール10^<-4>M刺激によるモチリン分泌抑制を検討した。その結果10^<-6>Mの4-DAMPによってモチリン分泌は完全に消失した。 3.ソマトスタチン、ボンベシンによる分泌反応今までにモチリンを分泌を調節するとin vivo実験にて報告されていたソマトスタチン、ボンベシンは、モチリン分泌を変化させなかった。従って、これらのホルモンによるモチリン分泌調節はモチリン産生細胞への直接作用とは考えられないことが明らかになった。以上の結果より、モチリン細胞は、細胞上に存在するM3レセプターを介しアセチルコリン刺激によって分泌が制御されている事が明らかになり、神経による消化管ホルモン分泌調節の新たな機序が想定された。
