肝毛細血管膜における脂質透過性を制御する因子に関する研究

研究概要	肝臓からの胆汁中への脂質分泌は肝毛細胆管膜を介して行われているが、肝内をvesicleとして輸送されてきた脂質が胆汁中へ放出されるためには、その第一段階として肝毛細胆管膜の内層に癒合しなければならない。そしてある種の輸送系を介して排泄されると考えられている。本年度の研究はその癒合過程に焦点を当てて遂行された。まず、市販のegg yolk phosphatydilcholine(EYPC),soybean PC,各種synthetic PCを用いてunilamellar liposomeを作製し、その凝集・融合態度の経時的観察からその物性の特徴を検討した。Vidio-enhanced differential contrast microscopyによる形態学的観察と、laser diffraction particle analyzerによるサイズ検索から、liposomeの融合性は構成するPCのアシル鎖脂肪酸の飽和度(逆相HPLCによって算出された疎水性強度)と、共存する中性ステロールの相対的濃度に依存することが明らかとなった。即ち、分子内炭素間の飽和度が高度であるほど、相互の凝集・融合性が強く、さらにwater-free spaceでのコレステロール分子配列が高度になると、凝集機転が機能しやすくなることが判明した。さらに、rhodamin Bをinsertされたliposomeを用いたfusion実験では、構成リン脂質の疎水性が強い程、fusibilityが高いことが判明した。今後、生体膜としてラット毛細胆管膜ヴェジクルを調整して、同様の現象が生理的に存在するかを確認しなければならない。
