冠血流調節における内皮由来弛緩因子の役割に関する研究:in vivoモデルによる研究

研究概要

冠血流量の調節における内皮由来一酸化窒素(EDNO)の役割を麻酔犬を用いたin vivoの実験系にて検討し、これまでに冠抵抗血管におけるトーヌスの調節にEDNOの基礎分泌が大きく関与していること(Arch int Pharmacodyn 1994;327:251-265)、一過性冠動脈閉塞に続く反応性充血にもEDNOが関与していること(Am J Physiol 1992;263:H8-H14、Cardiovascular Res 1996 in press)を報告した。またEDNO阻害時、心房ペーシングに伴う心筋酸素消費量増加時には心筋アデノシン放出の増加により代償されることを明らかにした(Am J Physiol 1996;270:H427-H434)。さらにニトローL-アルギニンメチルエステルの経口投与により慢性的にEDNOを抑制した麻酔犬において、心房ペーシングに伴う心筋酸素消費量増加時の冠血流増加、心筋収縮は対象群と差がなかったが、アデノシンレセプターブロッカーの8-フェニィルテオフィリン投与後は慢性阻害群において有意に冠血流増加、心筋収縮が抑制された。これより慢性的なEDNO抑制状態においては心筋酸素消費量増加時の代謝性冠血流調節にアデノシンの役割が重要である事が示された(第60回日本循環器学会総会 1996発表予定)。
