研究概要

甲状腺刺激ホルモン(TSH)受容体の細胞膜貫通領域をsite-directed mutagenesisにより変異させ、Cos-7細胞にトランスフェクションして表現実験を行い、非刺激下、TSH・TSAb(バセドウ病甲状腺刺激抗体)刺激下でのcAMP・イノシトール燐酸産生能とTSH結合能の測定およびTSH受容体の一部の合成ペプチドに対する特異抗体を用いたウエスタンブロットによる受容体蛋白の確認を各変異体についておこなった。その結果、第2細胞内ループの中央部(アミノ酸残基525-527;メチオノニンスタート部位を1とした番号)がGsとの相互作用に重要であることが明かとなった。また、Gqとの相互作用については以前報告していた第3細胞内ループの他、第1細胞内ループ全体、第2細胞内ループ、細胞内末尾部のN末側と広く分布していることを明かとした。さらに、細胞外ループについても同様の検討を行い、第3細胞外ループもcAMP系とイノシトール燐酸系の両方のシグナル伝達に重要であることを明かとした。プランマー病に見いだされたTSH受容体の第6細胞膜貫通ヘリックス部の4種類の点変異がいずれも刺激無しで活性化されている、いわゆるconstitutive activationを起こすものであることを表現実験によって示し、疾患の直接の原因となっていることを証明した。また、糖付加部位のミュータジェネシスにより、TSH受容体のアスパラギンリンクの糖鎖には種差があることを明かとした。
