研究概要	本研究では、Na^+/K^+-ATPase阻害剤(ウワバイン)とインスリンで前処置した血管平滑筋細胞(VSMC)を用いて、Myelin Basic Protein(MBP)のリン酸化を指標にしてMitogen-activated protein kinase(MAP)キナーゼを測定し、アルギニン・バゾプレシン(AVP)とアンジオテンシンII(AII)によるMAPキト-ゼ賦活化に対するウワバインとインスリンの効果を検討した。細胞内蛍光色素法により、細胞内遊離Ca^<2+>濃度([Ca^<2+>]i)と細胞内pH(pHi)を測定し、ウワバインとインスリンの効果の細胞内機序を解明した。(1)1mMウワバインでVSMCを数分間前処置すると、AVPとAIIによるMAPキナーゼ賦活化は増強された。また、同処置はCa^<2+> ionophore(ionomycin)とprotein kinase C(PKC)賦活剤(PMA)によるMAPキナーゼ活性化も増強された。1mMウワバインは、AVPとAII受容体には影響を与えなかったが、細胞内Na濃度([Na^+]i)の上昇による細胞膜電位の上昇とAVPとAIIによる脱分極を促進した。AVPとAIIによる[Ca^<2+>]i増員もウワバインは増大した。(2)1μMインスリンで24時間前処置すると、基礎値とAVPそしてAIIによるMAPキナーゼ賦活化が増強された。AVPのV_1受容体の親和性はインスリンにより増大し、AVPとAIIによる[Ca^<2+>]i増員もインスリン前処置により増強された。また、インスリンは、細胞内pHの基礎値を上昇させた。以上により、本能性高血圧症と糖尿病の動脈硬化の進展機序に血管収縮物質が関与することが示唆された。
