悪性骨軟部腫瘍の基底膜浸潤の分子機構の解析およびその抑制法の検討

研究概要	転移に際し、腫瘍細胞は、血管内皮下のバリヤ-である基底膜を浸潤破壊する必要があり、基底膜浸潤は、転移成立に極めて重要なステップと考えられている。平成6年度に、線維肉腫細胞においては、基底膜の酵素的破壊に働くtype IV collageneseの活性が、72kDa, 92kDaのtypeIV collagenaseとcollagenaseのインヒビターであるTIMPsの発現のバランスにより規定されており、細胞内cAMPを上昇させる薬剤(cAMPアナログなど)を投与すると、腫瘍細胞の、TIMP1、TIMP2の発現上昇によりtype IV collagenese活性が低下し、浸潤能、転移能が抑制されることを明らかにした。本年度は、更に、細胞内cAMPを上昇させる薬剤を投与により惹起される、TIMP1、TIMP2の発現上昇によるtype IV collagenese活性の低下、および浸潤能、転移能の抑制を、mRNAレベルだけでなく、蛋白レベルでも確認した。すなわち、72kDa, 92kDaのtyp IV collageneseとTIMPsの蛋白に対する抗体を遺伝子工学的手法により作製し、Western blottingにてcAMPアナログなどによりTIMPsの蛋白発現が上昇すること、また、TIMPsの中和抗体により、浸潤能が回復されることを見いだした。また、基底膜浸潤を果たし、標的臓器実質内に着床した腫瘍細胞は、既存の血管から新生血管を誘導し、酵素や栄養物を得て増殖していく。この腫瘍血管新生も転移形成に重要なプロセスだと考え、腫瘍血管新生の研究にも着手した。まず、規定膜抽出物マトリゲルを用いた新しいin vivo腫瘍血管新生定量法を開発した。本法を用いると、既存の血管新生阻害剤であるTNP70による血管新生阻害と腫瘍増殖抑制を確認することができた。
