移植腎浸潤細胞におけるT細胞抗原レセプターの解析

研究概要	移植腎浸潤T細胞の多様性を解析する手段として、T細胞レセプター可変域遺伝子(TCRV-gene)に特異的なprimersを用いるPCR法が広く行われている。しかしすべてのV genesをカバーするためには、1検体につき多数(Vα<30,Vβ<25)のPCRをセットする必要がある。そのため、バイオプシー試料などの微量な臨床材料を用いる際には十分な解析が不可能となる場合がある。研究代表者らは、anchored PCRにより増幅した検体のTCR-cDNAを膜固定化Vgeneとハイブリダイズさせることでこの問題を解決し、さらにnon-RI検出を組み合わせた方法を確立し、移植腎バイオプシー試料の解析に応用した。以下に方法の概略を示す。まず既知VgenesをすべてカバーするVα34種,Vβ28種について、V領域だけを含むplasmid clonesを調製し、そのDNA1μgずつをナイロン膜に固定した。移植腎バイオプシー試料および患者PBLからmRNAを分離し、合成したcDNAの上流にanchor oligonucleotideをligation後、biotin標識anchor primerおよびC-region primerによりTCR-cDNAを増幅・標識した。膜固定V遺伝子とハイブリダイズさせ、洗浄後、HRP-streptavidinを介して発色させ、492nm吸光度測定によりV usageを測定した。この解析法に必要な検体の量は抹消血約0.5ml相当、拒絶腎バイオプシー試料の1/2であった。本年度解析した腎移植例5例のうち4例において特定のV geneに偏りが見られ、特にそのうちの2例はPBLには見られない顕著な偏りを示し、特定の浸潤T細胞クローンによる拒絶反応惹起をうかがわせた。このように我々が開発した方法は、腎バイオプシー試料を用いたT細胞の解析に有用であることが実証された。
