哺乳類顎顔面領域におけるレチノイン酸催奇形機序の解明

研究概要

1 特異的な顎顔面奇形を誘発するレチノイン酸処理条件の確立
全胚培養下で胎齢9.0日および9.5日ラット胚子を2x10-7Mのレチノイン酸を添加した培養液中に6時間曝露し、その後無添加培養液に移して48時間培養すると、それぞれ特異的な顎顔面奇形が誘導された。前者の処理の場合は、第一咽頭弓のサイズが減少し、第二咽頭弓様の形態になり、後者の場合は第一及び第二咽頭弓が癒合した。
2 レチノイン酸処理胚子における頭部神経堤細胞の移動パターン
1の条件下でレチノイン酸処理を行ったのち、胚子の頭部神経堤細胞を生体為害性のない蛍光色素により標識し、全胚培養下で発生を進ませ、培養終了後、標識された細胞の移動経路を、蛍光顕微鏡により追跡し、正常発生の場合と比較検討した。胎齢9.0日ラット胚をレチノイン酸処理した場合、頭部神経堤細胞の移動パターンが正常とは異なり、第一咽頭弓へ移動する神経堤細胞が第二咽頭弓にも移動し、逆に本来第二咽頭弓へ移動するべき神経堤細胞が第一咽頭弓領域である三叉神経節に移動した。ことが明かになった。これに対し、胎齢9.5日ラット胚をレチノイン酸処理した場合は、正常発生における分節的な移動パターンは保持されていた。すなわち、本来第一咽頭弓へ移動する神経堤細胞は癒合した咽頭弓の前方領域のみを充填し、一方、第二咽頭弓へ移動すべき神経堤細胞は癒合した咽頭弓の後方領域のみを充填した。
以上より、レチノイン酸による顎顔面奇形の誘導がきわめて発生段階特異的であって、頭部神経堤細胞の挙動がこれに深く関わることが明らかになった。
