哺乳類顎顔面領域におけるレチノイン酸催奇形機序の解明‥先天異常のモデルとして

研究概要	昨年度は、特異的な顔面奇形を誘発するレチノイン酸処理条件(全胚培養下で胎齢9.0日および9.5日ラット胚子を2x10-7Mのレチノイン酸を6時間処理する)を確立した。前者の処理では、第一咽頭弓のサイズが減少し第二咽頭弓様の形態になり、後者では第一及び第二咽頭弓が癒合した。このレチノイン酸処理条件下で頭部神経堤細胞の移動経路を追跡したところ、正常発生胚子にみられる頭部神経堤細胞の分節的な移動が時期特異的に(9.0日胚子処理でのみ)阻害されることが明らかになった。本年度は以下のことについて正常発生胚子との比較を行い、レチノイン酸の効果がきわめて発生段階特異的であることをさらに明らかにした。 1)脳神経の形態の変化神経を特異的に染める抗体(2H3)によりwhole mount免疫染色を行い、脳神経の形態を観察した。胎齢9.0日ラット胚をレチノイン酸処理した場合、三叉神経の眼枝の形態は正常発胚子を変わらなかったが、上顎枝、および下顎枝は典型的な形態を失っていた。これに対し、胎齢9.5日ラット胚子を処理した場合は、咽頭弓の癒合とともに三叉神経節と顔面神経節も癒合することが観察されたが、三叉神経の形態自体は正常胚子のものと変わらなかった。 2)CRABP-I分子の局在の変化細胞質内でレチノイン酸と特異的に結合するCellular retinoic acid binding protein I(CRABP-I)タンパクに対する特異抗体を用いて分子の局在を組織学的に検索した。正常発生胚子ではCRABP-Iタンパクは第4-6ロンボメア(寮脳分節)および神経堤由来と考えられる第2-4咽頭弓間葉細胞に蓄積していた。胎齢9.0日ラット胚子をレチノイン酸処理した場合、CRABP-Iは正常発生胚子では局在性が低い第1咽頭弓間葉細胞や第1-3ロンボメアでも蓄積していた。これに対し、胎齢9.5日ラット胚子を処理した場合は、CRABP-Iの分布様式は正常発生胚子の場合と変わらなかった。
