歯周病原性細菌による骨吸収活性発現におけるサイトカインの役割

研究概要

歯周病原性細菌Actinobacillus actinomycetemcomitansから抽出精製した莢膜多糖をマウス骨髄細胞とともに8日間培養したところ、著しい数の破骨細胞形成が認められた。これまで我々が、構造を明らかにしたA.actinomycetemcomitans血清型a,b,cの莢膜多糖を比較検討したところ、血清型bの莢膜多糖にのみ強い活性発現が認められた。この結果は、A.actinomycetemcomitans血清型bが最も強い病原性を有するというこれまでの報告を支持するものであった。一方、破骨細胞形成に関与していると予想されるサイトカインに対する中和抗体を用いて、活性発現におよぼす影響を調べたところ、IL-1αに対する中和抗体で著しい阻害が認められた。プロスタグランデイン合成阻害剤であるインドメサシンを培養系に加えたところ、強い阻害効果が認められた。さらに、マウス骨髄細胞培養上清中のサイトカイン量をELISA法で調べたところ多量のIL-1αが検出された。プロスタグランデインの定量も既存のRIA法で行なったところ、莢膜多糖の刺激により有意のプロスタグランデインの産生が認められた。以上の結果から、歯周病原性細菌A.actinomycetemcomitansの莢膜多糖による破骨細胞の活性化および分化誘導にはIL-1αとプロスタグランデインが深く関与していることが明らかとなった。さらに、歯周病で見られる歯槽骨吸収では、宿主側の要因が関与している可能性が強く示唆された。
