高等動物におけるDNAのメチル化と脱メチル化に関する研究

研究概要

本研究計画は、高等動物の染色体DNAのメチル化状態を規定する因子の一つであるDNAメチルトランスフェラーゼ(MTase)とメチル化の機能およびメチル化状態の調節に関わるタンパク質について理解を深めることを目的とする。研究期間、平成6年度から7年度にかけて以下のような成果が得られた。
1.ニワトリ(鳥類)とXenopus laevis(両生類)のMTaseのcDNAを単離してその塩基配列を決定した。また、cDNAを発現ベクターにいれて培養細胞で発現させその性質を調べた。強制発現させたマウスのMTaseはニワトリのMTaseよりも高いde novoメチル化活性を示した。
2.プロトオンコジーンc-Mycが結合するDNAモチーフをメチル化したとき特異的に結合するDNA結合タンパク質、MMBP-3を見つけた。このMMBP-3は細胞が増殖しているときに活性が高く、細胞の増殖を抑制するとその結合活性は低下した。
3.筋芽細胞にMTaseを強制発現させると、筋管形成の終末分化が促進された。これは筋芽細胞が増殖しているときからMyoDの発現が促進していたためであり、このMyoDの発現促進は転写活性の上昇によるものであった。このときMyoD遺伝子の転写開始付近の特定のCpG配列がメチル化されていた。
4.培養繊維芽細胞を脱メチル化剤で処理したときに誘導される新しい遺伝子AZ1のcDNAを単離して構造を決定した。遺伝子のコードするAZ1タンパク質は精子細胞のアクロソーム前駆体内に局在した。
