定量的免疫電顕法を用いたシナプス小胞の前シナプス形質膜への開口分泌機構の解析-低分子量GTP結合蛋白質の作用機構を中心として-

研究概要	神経伝達において、シナプス小胞は前シナプス形質膜上の特定の位置であるCa^<2+>チャネル集合部位(アクティブゾーン)へと選択的に輸送され、前シナプス形質膜に接着したあと、Ca^<2+>流入後0.2ミリ秒以内という超高速度でそれと融合し、神経伝達物質を放出する。この超高速度開口分泌において、低分子量GTP結合蛋白質Rab3A/smg p25Aが、シナプス小胞の前シナプス形質膜への選択的輸送・接着・融合の段階に作用することを近年私共が見いだした。今年度の研究で、私はRab3Aのシナプス内小胞輸送における作用機構を明かにする目的で、Rab3Aの機能を調節する蛋白質を検索し、GTP結合型Rab3Aとのみ結合する標的蛋白質Rabphilin-3Aを同定した。さらに、シナプス内の局在を定量的免疫電顕法(プレエンベッティング1nm金粒子銀増感法)で解析した結果、Rabphilin-3AはRab3Aと同様にシナプス小胞に局在し、1個のシナプス小胞上にRabphilin-3Aが2分子、Rab3Aが6分子存在することが判明した。この結果から、Rab3Aは、シナプス小胞上でRabphilin-3Aと複合体を形成して機能していることが示唆された。私共の見いだしたこれらの事実は海外で広く確認され、世界的な分子生物学の教科書であるMolecular Biology of the Cellの13章PP643-645に、Rab3AがCa^<2+>依存性開口分泌に必須の因子であり、またRabファミリー低分子量GTP結合蛋白質が細胞内で小胞の輸送目的地の特異性の制御に関与していると引用された。さらに最近私は、神経発生や再生の際、神経突起の遠位端に位置し、シナプスの前駆体として標的細胞の識別に重要な働きをする成長円錐における小胞輸送制御機構に研究を進めた。成長円錐においても、Rab3A-Rabphilin-3Aが機能しており、しかも開口分泌が活動電位に依存せず、成長円錐形質膜上の細胞接着分子と細胞外基質・標的細胞との接着に依存する可能性が示唆された。
