自己免疫性脳脊髄炎と神経移植におけるサイトカインの役割

研究概要

中枢神経系での病変修復過程において、損傷部位から分泌されたサイトカインのネットワークを検討した。サイトカインの蛋白やメッセンジャーRNAは発現量が微量のことが多いので、従来のWestern blottingやNorthern blottingによる検索では十分とは言えなかった。本研究ではCompetitive PCRによってサイトカイン遺伝子を増幅し、かつ定量的に検出した。実験系としてはEAEと神経移植を用いた。
自己免疫性脳脊髄炎(EAE)においては炎症性サイトカイン(TNF-α、IFN-γ)と抑制性サイトカインIL-10が発症早期にピークを示し、EAE極期から回復期に速やかに減少した。もう一つの抑制性サイトカインTGF-β1は正常状態でも発現を認めEAEの臨床症状に平行して増減した。一方、移植片の拒絶過程では炎症性サイトカインに加えてcytolytic protein(perforin、granzyme B)が強く発現し、IL-10の発現が弱かった。これらのサイトカインの複雑なネットワークが中枢神経系の病変形成と炎症の収束に関与していると考えられた。
