メタンフェタミン逆耐性状態での中枢ガンマアミノ酪酸神経活動に対する検討

研究概要	申請者はこれまでメタンフェタミン逆耐性形成ラット脳内ドパミン(DA)神経系の変化に関して、DAの再取り込み機構の亢進、DA放出調節自己受容体の機能低下、DA放出調節を行うオピオイド受容体の中で抑制的に働くカッパ受容体の機能低下、及び促進的に働くミュ受容体の機能亢進が生じている結果を得て、これらのことから逆耐性形成ラットの脳内DA神経活動が亢進している状態が考えられる。このDA神経活動亢進が後シナプスDA受容体にどのように影響するか、その受容体が存在する神経細胞の活動にどのように影響するかに関しての神経化学的知見は少ない。そこで、DA神経がシナプスを形成している側坐核内GABA神経に注目し、この神経からのGABAの放出に及ぼすDA受容体の影響を調べた。方法の詳細は、交付申請書に記載したように脳内透析灌流法を用いたラット側坐核GABA細胞外濃度を経時的に測定し、DA受容体の中でD_1及びD_2受容体が側坐核GABA放出にどのように作用しているかを調べDA神経の神経伝達がGABA神経系に及ぼす効果を検討した。加えて、逆耐性形成ラットでのDA受容体のGABA放出への効果を対照群との間で比較検討し、その病態における神経伝達機構の変化を調べた。これまでに以下のような結果が得られた。側坐核細胞外GABA濃度は、高カリウム刺激によって上昇し、テトロドトキシン感受性が認められ、放出された伝達物質として考えることが可能である。D_1及びD_2受容体による側坐核GABA放出への効果は、D_1受容体拮抗薬局所灌流でGABA放出の抑制が、D_2受容体拮抗薬の局所灌流によるGABA放出の促進効果の減弱が認められた。これらのことは、側坐核でのGABA放出に対してD_1受容体は促進的に、D_2受容体受容体は抑制的に作用しており、逆耐性形成ラットでのGABA放出調節D_2E2の機能低下で出現することが考えられる。本研究で、申請期間内に検討できなかった部分について今後検討を続けて行く予定である。
